Recently, I learned from Shanghai Jiaotong University that the State Key Laboratory of Microbial Metabolism has made a breakthrough in the modification of DNA phosphorylation by the Delin research team. Relevant results were published online in the journal Nature-Communication.
DNA is the genetic material of life and consists of five elements: carbon, hydrogen, oxygen, nitrogen and phosphorus. In the previous work, the team led by Academician Deng Zixin of the Chinese Academy of Sciences discovered a new modification on DNA - phosphorylthiolation. This phenomenon is recognized by international peers as "the first natural modification found on the DNA backbone so far" and the sixth element currently found on the DNA backbone.
The Delin team used Escherichia coli B7A as the research object to establish two kinds of cutting-edge technologies, such as single-molecule real-time sequencing and iodine-cleavage-dependent deep sequencing. It is understood that the two sequencing methods complement each other, and the modified site has a conservative GpsAAC/GpsTTC short sequence characteristic. A DNA phosphorylation modification-restriction system exists in E. coli, and only part of the GAAC/GTTC site in the genome is recognized by the DNA phosphorylation modification enzyme. This finding suggests that the DNA phosphorylation modification-restriction system differs from the classical methylation modification-restriction system in that it has a special mechanism of action.
